Asparaginase
L-ASPARAGINASEObserving the qualitative difference observed between normal cells and those from childhood lymphoblastic leukemia - the leukemia cells are deficient in L-aspargine synthetase and depend on the supply of L-asparagine from the medium. The enzyme L-asparaginase (from E. coli) degrades L-asparagine to L-aspartic acid; depriving leukemic cells of an essential metabolite - may cause cell death. Indications: Acute leukaemia. Malignant lymphoma.. Dosage of L-Asparaginase50-200 k.u/kg body wt. daily or every alternate day by i.v. infusion. Contra-Indications: Hypersensitivity. Special Precautions of L-AsparaginaseAnaphylaxis can occur, allergic reaction (being a foreign protein). Paediatrics -- Safe. Pregnancy -- Should not be used. Lactation -- Should not be used. Elderly -- Use with caution. Side effects of L-Asparaginase
Drug Interactions of L-AsparaginaseVincristine and prednisolone increases the toxicity, diminishes or abolishes the effect of methotrexate on malignant cells.
