L-ASPARAGINASE

Observing the qualitative difference observed between normal cells and those from childhood lymphoblastic leukemia - the leukemia cells are deficient in L-aspargine synthetase and depend on the supply of L-asparagine from the medium. The enzyme L-asparaginase (from E. coli) degrades L-asparagine to L-aspartic acid; depriving leukemic cells of an essential metabolite - may cause cell death.

Indications: Acute leukaemia. Malignant lymphoma..

Dosage of L-Asparaginase

50-200 k.u/kg body wt. daily or every alternate day by i.v. infusion.

Contra-Indications: Hypersensitivity.

Special Precautions of L-Asparaginase

Anaphylaxis can occur, allergic reaction (being a foreign protein).

Paediatrics -- Safe.

Pregnancy -- Should not be used.

Lactation -- Should not be used.

Elderly -- Use with caution.

Side effects of L-Asparaginase

• Liver damage,
• pancreatitis and C.N.S symptoms (due to defective protein synthesis)

Drug Interactions of L-Asparaginase

Vincristine and prednisolone increases the toxicity, diminishes or abolishes the effect of methotrexate on malignant cells.

Cancer Tip Conventional cancer treatments, such as chemotherapy and radiation therapy, cannot distinguish between cancer cells and healthy cells. Consequently, healthy cells are commonly damaged in the process of treating the cancer, which results in side effects.
